Objectives: In this study, we investigate the anti-oxidant potential of herbal mix extract of Onion Juice, Ocimum Basilicum, orange peel, Cinnamomum Zeylanicum, ginger, Citrullus lanatus, Citrullus Vulgaris and Carrot seed)on testis structure and sperm parameters of rats.

Materials and methods: In this research, 30 male Wistar rats were segregated in 3 groups:1- Control, 2-healthy rats treated with 10 mg/kg of the herbal mix, 3- rats treated with 15 mg/kg of the herbal mix. Also, treatment groups received herbal mix for 8 weeks. After treatment period all of the animals were anesthetized, their blood sample were taken, the serum level of sexual hormones was measured then the testicles and epididymis were removed and sperm parameters, oxidative stress markers and expression of SF1 and LHCGR genes were assessed.

Results: Treatment groups with herbal mix increased the Johnson score and diameter of Seminiferous tubules as well as reduced the MDA plasma levels (p=0.001) and increased the SOD and GPX and sexual hormones (LH, FSH, and Testosterone) levels (p=0.001). Moreover, the expression of SF1 and LHCGR were notably increased in treatment groups. the sperm quality enhanced in the therapy groups (p<0.05).

Conclusion: The results of this study confirmed that the antioxidant role of herbal mix was improved the testicular structure and regulate the markers of oxidative stress and also enhanced the expression of SF1 and LHCGR as well as it can enhance the sperm quality.
