Objectives: It is well known that thyroid malfunction has a negative impact on male reproductive system. Besides, there is evidence indicating that extracellular matrix (ECM) components involves in the testicular development and function. In hypothyroid state marked disruption in the architecture of ECM in various tissues of mammals has been shown. As well as, significant changes in the germ cell population have been reported under hypothyroidism. Therefore, this study was designed to investigate the relationship between ECM proteins and germ cells apoptosis following hypothyroidism.
Materials and Methods: In this experimental study, 20 male Balb/C mice were divided into control group and hypothyroid group that was received 0.05% 6-n-propyl-2-thiouracil (PTU) through drinking water for 35 days. After confirmation of hypothyroidism, Real-Time PCR, immunohistochemistry, Periodic Acid-Schiff (PAS) staining, TUNEL assay and biochemical measurements were carried out.
Results: Laminin α5 and collagen IV mRNA levels were upregulated in the hypothyroid group (p<0.05). Moreover, a strong immunoreactivity of collagen IV and laminin α5 was observed in the basement membrane (BM) of hypothyroid mice (p<0.05). However, no significant difference was found in PAS staining between two groups. Furthermore, the mean number of TUNEL-positive germ cells increased significantly in the hypothyroid mice in comparison with the control ones. Also, PTU administration reduced superoxide dismutase (SOD) activity while increased malondialdehyde (MDA) level (p<0.05).
Conclusions: The data suggest that hypothyroidism may through induction of lipid peroxidation (LPx) and reduction in the antioxidant defense molecules activity lead to alterations in BM components and increased germ cells apoptosis.
